📚 IGCSE AQA Biology: Common Mistakes and Key Explanations | IGCSE AQA

In IGCSE AQA Biology, many students lose marks not because they don’t know the material, but because they make common errors in understanding key concepts or in applying their knowledge to exam questions. This article highlights frequent mistakes and provides clear explanations to help you avoid them.

1. Confusing Diffusion and Osmosis |

A very common error is mixing up diffusion and osmosis. Students often state that ‘osmosis is the movement of water from a low to a high concentration of solutes’.

In fact, osmosis is the net movement of water molecules from a region of higher water potential to a region of lower water potential through a partially permeable membrane. Water potential depends on both solute concentration and pressure. Pure water has the highest water potential (0 kPa); adding solutes lowers it. Therefore, water moves from a dilute solution to a more concentrated one across the membrane.

Another mistake is forgetting the membrane requirement. Diffusion can happen without a membrane, but osmosis always requires a partially permeable membrane. Students also lose marks by omitting the term ‘water potential’ in written answers.

Diffusion Osmosis
Net movement of particles from high to low concentration Net movement of water from high to low water potential
No membrane required Requires a partially permeable membrane
Substances can be gas or solute Only water

When answering exam questions, always specify ‘down the concentration gradient’ for diffusion and ‘from high to low water potential through a partially permeable membrane’ for osmosis.

2. Enzyme Denaturation and Temperature |

Many students think that enzymes are ‘killed’ at high temperatures or that denaturation means the enzyme is dead. This is a critical error.

Enzymes are proteins, not living organisms. High temperatures break the hydrogen bonds and ionic interactions that maintain the specific tertiary structure of the active site. This changes the shape of the active site so that the substrate can no longer fit. The enzyme is said to be denatured, and this change is usually irreversible. It is not death; the protein simply loses its catalytic function.

A related misconception is that any deviation from the optimum temperature ‘kills’ enzymes. In reality, lowering the temperature only reduces kinetic energy, leading to fewer successful collisions between enzyme and substrate, so the rate of reaction decreases. The enzyme remains functional and will regain activity when the temperature returns to optimum.

Students often fail to describe the effect of temperature on enzyme activity in a sequence: as temperature rises, kinetic energy increases, more enzyme–substrate complexes form, and the rate increases. At the optimum, it peaks. Beyond the optimum, heat causes denaturation, the active site changes shape, and the rate drops sharply.

When writing about pH, the same principle applies: extreme pH disrupts the bonds maintaining the active site shape, leading to denaturation. Avoid saying the enzyme ‘dies’.

3. Limiting Factors of Photosynthesis |

A classic error appears when interpreting graphs of light intensity and photosynthesis rate. Students claim that ‘photosynthesis stops increasing because the plant has enough light’.

The correct interpretation is that another factor is now limiting the rate. Initially, light intensity is the limiting factor; increasing it increases the rate. When the curve levels off, something else, such as CO₂ concentration or temperature, has become the limiting factor. At very low light intensity, CO₂ levels may be high, but if light is insufficient, light is still the limiting factor.

Students commonly mix up the roles of temperature and CO₂. At low CO₂ concentration, Rubisco cannot fix much carbon, and the rate is low even if light and temperature are optimal. Temperature affects enzyme activity, particularly in the Calvin cycle. At excessive temperatures, enzymes such as Rubisco can denature, and stomata may close, reducing CO₂ intake, which then limits photosynthesis.

Another common pitfall is drawing or describing the graph incorrectly, such as not starting the line at the origin or missing the plateau. Make sure you can explain what ‘limiting factor’ means and that you refer to specific experimental conditions.

4. Aerobic and Anaerobic Respiration |

One of the most frequent mistakes is confusing the products of anaerobic respiration in humans with those in yeast.

In human muscle cells during vigorous exercise, anaerobic respiration produces lactic acid (C₃H₆O₃) and a small amount of ATP. No ethanol or CO₂ is formed. The equation can be summarised: C₆H₁₂O₆ → 2C₃H₆O₃. In yeast and many plants, anaerobic respiration produces ethanol (C₂H₅OH) and carbon dioxide: C₆H₁₂O₆ → 2C₂H₅OH + 2CO₂.

Many students write that anaerobic respiration produces no energy. In truth, it releases only a small amount of energy (2 ATP molecules per glucose) compared to aerobic respiration (up to about 32 ATP per glucose). The statement ‘no energy’ is incorrect and will lose marks.

Oxygen debt is another source of error. Students often say that extra oxygen is breathed in to ‘break down lactic acid in the muscles’. The truth is that most lactic acid is transported via the blood to the liver, where it is oxidised back to pyruvate or converted to glucose using oxygen. The increased breathing rate after exercise repays the oxygen needed for this process.

Always specify that aerobic respiration requires oxygen and produces carbon dioxide and water, while releasing a large amount of energy. Use correct equations and avoid mixing up organisms.

5. Genetics: Genotype, Phenotype and Probability |

Students often cannot distinguish between homozygous and heterozygous, or they think a heterozygous individual shows a blend of the two alleles. In Mendelian inheritance, the dominant allele is expressed in the phenotype even when only one copy is present.

Another mistake is using different letters for alleles, e.g. ‘B’ for brown eyes and ‘b’ for blue eyes is correct, but ‘B’ and ‘e’ is wrong. Always use the same letter with upper case for dominant and lower case for recessive.

When constructing Punnett squares, some students confuse the probability for a single child with the ratio in a large number of offspring. If two heterozygotes (Rr) reproduce, the probability of a child showing the recessive trait is 1/4, but that does not mean the fourth child will necessarily have it. Each child has the same 25 % chance. Also, when the question states that a child already exhibits a dominant phenotype, the probability that the child is a carrier (heterozygous) is 2/3, not 1/2, if both parents are heterozygous. This subtle point is frequently tested.

Additionally, remember that the phenotype is the result of genotype interacting with the environment. For instance, plant height can be influenced by light and nutrients, not just genes. In AQA exams, always define genotype, phenotype, homozygous, heterozygous, dominant, and recessive clearly.

6. Natural Selection and Evolution Misconceptions |

A dangerously common error is phrasing evolution as though organisms deliberately change. Phrases like ‘bacteria develop antibiotic resistance in order to survive’ or ‘giraffes stretched their necks to reach leaves’ imply intentional adaptation, which is wrong.

Correct thinking: Within a population, there is random genetic variation. By chance, some bacteria have a mutation that makes them less susceptible to an antibiotic. When the antibiotic is applied, susceptible bacteria die, while resistant ones survive and reproduce. Over generations, the frequency of the resistance allele increases. The environment acts as a selection pressure, not a designer.

Students also confuse ‘survival of the fittest’ with ‘survival of the strongest’. Fitness in biology means the ability to survive and reproduce in a specific environment. A smaller, quicker organism might be fitter than a larger, stronger one. Natural selection does not produce perfection; it works with available variation.

When explaining MRSA or antibiotic resistance, provide a step-by-step sequence: random mutation, variation, non-competitive advantage when antibiotic present, survival, reproduction, increase in proportion of resistant bacteria. Avoid any language implying bacteria ‘learn’ or ‘get used to’ drugs.

7. Energy Transfer in Food Chains |

Many students incorrectly assume that all energy from one trophic level is passed on to the next. In reality, only about 10 % of the energy is transferred; the rest is lost.

The energy ‘lost’ is not destroyed but is released to the environment as heat during respiration, or remains in uneaten parts, undigested material excreted as faeces, and in waste products like urea. Students often write ‘energy disappears’, which is unacceptable in a scientific answer.

Because energy is lost at each level, food
