Cloning is the production of genetically identical copies of an organism or individual cells. In GCSE OCR Biology, you must be able to describe natural and artificial cloning methods in both plants and animals, explain the steps involved in tissue culture and adult cell cloning, and discuss the ethical and practical implications of cloning technology.

1. What is Cloning? | 什么是克隆?

A clone is a group of genetically identical cells or individuals that originate from a single ancestor. Cloning is a form of asexual reproduction, because it does not involve the fusion of gametes. The offspring inherit all their DNA from one parent and are exact genetic copies.

2. Natural Cloning in Plants | 植物自然克隆

Many plants reproduce asexually through natural cloning. For example, strawberries produce runners – horizontal stems that grow along the soil surface and form new plantlets at their tips. Potatoes form underground tubers, each of which contains buds that can grow into a new plant. Onions and garlic use bulbs, which are short underground stems surrounded by fleshy leaf bases that store food and can divide to generate new bulbs.

3. Natural Cloning in Animals | 动物自然克隆

Natural cloning is less common in animals, but some invertebrates reproduce in this way. Hydra, a small freshwater organism, produces buds that pinch off and develop into independent, genetically identical polyps. Aphids can reproduce by parthenogenesis in summer, where females produce live, female offspring without mating. These offspring are clones of the mother.

4. Artificial Cloning in Plants: Cuttings | 植物人工克隆:插条

Gardeners and farmers have long used cuttings to produce new plants that are genetically identical to a parent with desirable traits. A cutting is a piece of stem or leaf taken from a healthy plant and placed in moist soil or water. The cut end develops roots, and eventually a new plant grows. To prevent infection, the cutting is often dipped in rooting powder containing auxin, a plant hormone that stimulates root growth.

5. Tissue Culture and Micropropagation | 组织培养与微繁殖

Tissue culture, also called micropropagation, is a modern method of cloning plants under sterile conditions on a nutrient-rich gel. It starts with a tiny piece of plant tissue called an explant, which can be taken from a meristem (growth tip). Because meristem cells are often virus-free, micropropagation can produce large numbers of disease-free plants quickly.

6. Micropropagation Steps | 微繁殖步骤

Step 1: A small piece of tissue (explant) is taken from the parent plant, typically from the shoot tip or meristem.

Step 2: The explant is sterilised using bleach or alcohol to kill any bacteria or fungi.

Step 3: The sterile explant is placed on an agar medium containing nutrients, sugars and a carefully balanced mixture of plant hormones (auxins and cytokinins) that stimulate cell division.

Step 4: The cells divide and form a mass of undifferentiated cells called a callus. This callus can be subdivided to encourage further growth.

Step 5: By adjusting the hormone balance, the callus differentiates into small shoots and roots, forming tiny plantlets.

Step 6: The plantlets are transferred to soil or compost in a greenhouse, where they harden off and grow into mature plants that are all clones of the original parent.

7. Animal Cloning: Embryo Transplants | 动物克隆:胚胎移植

Embryo transplantation is a method used on animals, particularly high-value cattle. A prize female is treated with hormones to produce many eggs, which are then fertilised in vitro using sperm from a desirable male. The resulting fertilised eggs develop into early embryos composed of a few identical cells.

This technique allows a farmer to produce many offspring from the best parents in a short time.

Adult cell cloning, also known as somatic cell nuclear transfer (SCNT), produces a clone by transferring the nucleus from an adult body cell into an enucleated egg cell. The nucleus is removed from an unfertilised egg cell (enucleation), leaving an empty egg with all the cytoplasmic machinery needed for development.

成体细胞克隆,也称为体细胞核移植(SCNT),通过将一个成年体细胞的细胞核移植到去核卵细胞中来产生克隆。从未受精卵细胞中取出细胞核(去核),留下一个含有发育所需所有细胞质成分的空卵。

A complete nucleus from a body cell, such as a skin or udder cell, is inserted into the enucleated egg. A mild electric shock stimulates the egg to begin dividing, as if it had been fertilised. The resulting embryo is allowed to grow into a blastocyst and is then implanted into the uterus of a surrogate female. The offspring is genetically identical to the organism that supplied the nucleus.

Dolly the sheep, born in 1996, was the first mammal to be cloned from an adult somatic cell. Scientists at the Roslin Institute took a nucleus from a mammary gland cell of a Finn Dorset ewe and transferred it into an enucleated egg cell from a Scottish Blackface ewe. After the electric pulse, the embryo was placed into a surrogate Scottish Blackface. Dolly was genetically identical to the Finn Dorset ewe.

However, Dolly developed arthritis and lung disease at a relatively early age and lived only until about six years, raising concerns about the health of cloned animals.

Cloning enables the rapid multiplication of organisms with desirable traits. In agriculture, farmers can produce whole herds of high milk-yielding cows or disease-resistant crops without waiting for sexual reproduction cycles. Rare or endangered species can potentially be saved by cloning individuals, though genetic diversity remains a concern.

Cloning techniques often have a low success rate. Many cloned embryos fail to implant or develop abnormalities. Cloned animals may suffer from large offspring syndrome, premature ageing and weakened immune systems. Dolly’s early health problems illustrate these risks.

There are also significant ethical objections. Many people believe that cloning animals is unnatural and raises animal welfare issues, especially due to the high number of failed pregnancies and malformed offspring. The prospect of human cloning is widely condemned. There is also a concern that cloning reduces genetic variation, which could make populations more vulnerable to diseases or environmental changes.

In the exam, be precise when describing the steps of micropropagation and adult cell cloning. Use terms such as explant, callus, aseptic technique, enucleation and surrogate mother. Many students confuse embryo cloning with adult cell cloning; remember that embryo splitting uses cells from a developing embryo, whereas SCNT starts with a nucleus from an adult body cell.
