Chromatography is a powerful separation technique used to separate and identify components in a mixture. It is widely applied in forensic science, food analysis, and pharmaceuticals. For IGCSE Chemistry, you need to understand the principles, interpret chromatograms, and calculate Rf values accurately.

1. Introduction to Chromatography | 色谱法简介

Chromatography is a method for separating dissolved substances that move at different rates through a stationary phase when carried by a mobile phase. The term originated from the Greek words for ‘colour writing’, as early experiments separated plant pigments.

2. Principles of Chromatography | 色谱法原理

It is important to use a pencil for the baseline, as ink would dissolve in the solvent and interfere with results. The paper must not touch the sides of the container, and the solvent level must be below the baseline.

5. Thin-Layer Chromatography (TLC) | 薄层色谱法

After the solvent front has reached a suitable height, the plate is removed and dried. Spots are often visualised under UV light or with a locating agent if they are colourless.

6. Interpreting Chromatograms | 色谱图解析

A chromatogram is the visual result showing separated spots at different heights. The number of spots indicates the minimum number of components in the mixture. Identical substances will rise to the same height under the same conditions.

The retention factor (Rf) is a ratio used to identify a substance under consistent conditions. The formula is:

Rf = distance moved by substance / distance moved by solvent front

Both distances are measured from the baseline to the centre of the spot and to the solvent front. Rf has no units and is always less than 1. It is constant for a given substance in a given solvent and temperature.

A frequent mistake is measuring distances from the bottom of the paper instead of the baseline. Always use the pencil line. Also, make sure the spots are small and not overlapping on the baseline.
