📚 Cloning in IGCSE Edexcel Biology | IGCSE Edexcel

Cloning is the process of producing genetically identical copies of an organism, cell, or DNA fragment. For IGCSE Edexcel Biology, you need to understand both natural and artificial cloning methods, their applications in plants and animals, and the ethical considerations involved. This article covers the key points you must know for the exam, including micropropagation, embryo transplants, and adult cell cloning, together with the advantages and disadvantages of each technique.

1. What is Cloning? Definition and Natural Cloning |

Cloning produces individuals with identical genetic material. In nature, asexual reproduction leads to clones, as seen in bacteria, some plants (runners, bulbs, tubers), and identical twins in animals. Natural clones are the basis for many agricultural practices, such as taking cuttings from a desirable plant.

Identical twins occur when a fertilised egg splits into two embryos, each with the same DNA. In plants, structures like strawberry runners and potato tubers grow into genetically identical new plants. These are all examples of natural cloning, where no human technology is involved.

2. Artificial Plant Cloning: Cuttings and Tissue Culture |

Gardeners have used cuttings for centuries. A stem, leaf, or root is cut from a parent plant and placed in moist soil or water. Auxin hormones encourage root development, and the cutting grows into a clone of the parent. This method is cheap and simple but can fail due to infection or poor conditions.

Modern plant cloning often uses micropropagation (tissue culture). Small pieces of plant tissue (explants) are sterilised and placed on a nutrient agar medium containing hormones. Cells divide and form a callus, which then differentiates into tiny plantlets. These are transferred to soil once they have roots and shoots. Micropropagation produces thousands of identical plants quickly and under disease‑free conditions.

  • Advantages of micropropagation: rapid multiplication, disease‑free plants, can preserve rare species. /
  • Disadvantages: expensive, requires sterile lab, all plants are equally susceptible to a new disease. /

3. Animal Cloning: Embryo Transplants |

Embryo transplants are a traditional form of animal cloning used to increase the number of offspring from a prized female. The process involves giving a donor cow hormones to produce many eggs, which are then fertilised artificially by sperm from a top‑quality bull. The developing embryos are flushed out, split, and implanted into surrogate mothers. Because the embryos came from the same parents, the resulting calves are clones of each other, though not of the parents.

This technique is widely accepted in farming. It allows a farmer to produce several genetically identical female calves with the mother’s desirable milk yield and the father’s good beef qualities, without waiting for the cow’s natural cycle. The embryos can be frozen and stored for later use. However, it reduces genetic diversity within a herd.

4. Adult Cell Cloning: Somatic Cell Nuclear Transfer (SCNT) |

Adult cell cloning, also known as somatic cell nuclear transfer, was made famous by Dolly the sheep in 1996. The nucleus is removed from an unfertilised egg cell (enucleation) and replaced with the nucleus from a body cell of the animal to be cloned. An electric shock stimulates the egg to begin dividing, forming an embryo. This embryo is then implanted into a surrogate mother. The offspring is a clone of the nucleus donor.

The key steps are: 1) Remove nucleus from an egg cell; 2) Take a diploid nucleus from an adult donor cell; 3) Insert the diploid nucleus into the enucleated egg; 4) Give an electric shock to trigger mitosis; 5) Culture the embryo in a lab; 6) Implant into a surrogate’s uterus. The resulting clone is genetically identical to the donor of the body cell, not the egg donor or surrogate.

5. Comparison of Embryo Transplants and Adult Cell Cloning |

Source of genetic material / 遗传物质来源 Sexual reproduction (sperm + egg) / 有性繁殖 Diploid nucleus from a body cell / 体细胞的二倍体核
Offspring are clones of each other, not parents / 后代彼此克隆,非父母克隆 Clone is identical to the body cell donor / 克隆体与体细胞供体相同
Number of cloned individuals / 克隆个体数量 Multiple, from one fertilisation / 一次受精可产生多个 Typically one at a time / 通常每次一个

Because adult cell cloning bypasses sexual reproduction, it raises more ethical concerns. However, it allows a copy of a specific adult animal with known traits, whereas embryo transplants only multiply embryos from selected parents.

6. Advantages of Cloning |

Cloning can preserve endangered species by increasing population numbers without requiring breeding pairs. It enables mass production of organisms with useful traits: high‑yield crops, disease‑resistant plants, or animals that produce pharmaceutical proteins in their milk (pharming). In medicine, cloned embryonic stem cells could be used for therapeutic purposes, such as replacing damaged tissues.

In agriculture, cloned plants guarantee uniformity of fruit size, ripening time, and pest resistance, which simplifies harvesting and increases profit. Genetically identical animals can help scientists study diseases by removing genetic variability as a confounding factor.

7. Disadvantages and Risks of Cloning |

Cloning reduces genetic variation, making populations vulnerable to diseases or environmental changes – a single threat could wipe out an entire genetically uniform crop. Cloned animals often suffer from health problems due to incomplete reprogramming of the adult nucleus, leading to premature ageing and organ failure. Dolly lived only half the normal lifespan of a sheep.

Ethical arguments against cloning include: it is seen as ‘playing God’ by some; animals may suffer; the technique could be misused for human reproductive cloning, which is illegal in most countries. The high cost and low success rate also limit its practical use.

8. Tissue Culture and Micropropagation in More Detail |

Micropropagation relies on the fact that plant cells are totipotent – each cell can develop into a whole plant given the right conditions. The nutrient medium contains macro‑ and micronutrients, sugars for energy, and plant hormones such as auxins and cytokinins. The ratio of these hormones determines whether cells form roots or shoots.

The process must be carried out under aseptic conditions to prevent bacterial or fungal contamination, which would kill the explants. All equipment and media are sterilised, and work is done in a laminar flow hood. Once plantlets have developed, they are gradually acclimatised to normal atmospheric conditions before being planted out.

9. Commercial Uses of Plant Cloning |

Orchids and other ornamental plants are routinely propagated by micropropagation to meet market demand. Growers can produce thousands of identical flowers in a controlled way, ensuring colour, shape, and flowering time match customer expectations. Banana plants, which are sterile and do not produce viable seeds, are cloned through tissue culture to supply the global market.

Crop plants such as potatoes can be mass‑propagated virus‑free using meristem tissue culture. The meristem is taken from the plant tip and is often virus‑free even in an infected plant. This method helps maintain healthy seed stock.

10. Therapeutic Cloning and its Potential |

Therapeutic cloning uses the same SCNT technique to produce an embryo, but the embryo is never implanted into a uterus. Instead, embryonic stem cells are harvested from the blastocyst after a few days. These stem cells are pluripotent – they can differentiate into any cell type – and could be used to grow replacement tissues or organs, genetically matched to the patient to avoid immune rejection.

This technology is still experimental and faces strong ethical opposition because it involves creating and destroying an embryo. Many people distinguish between reproductive cloning, which aims to create a new individual, and therapeutic cloning, which aims only to produce cells for treatment. In exam answers, it is crucial to clarify which type you are discussing.

11. Cloning vs Genetic Engineering: Know the Difference |

Cloning produces genetically identical copies of an existing organism or DNA fragment, without altering the DNA sequence. Genetic engineering, by contrast, alters the genetic makeup by inserting, deleting, or modifying genes. A popular exam question asks you to compare the two: for example, genetically modified bacteria producing insulin involve genetic engineering, while micropropagation of a GM plant produces clones of the modified plant.

In the context of animals, Dolly was not genetically modified; she was a clone of her nucleus donor. However, techniques can be combined – for instance, a transgenic animal (genetically modified) could be cloned to produce a herd of animals all carrying the desired transgene.

12. Exam Tips for Cloning Questions |

IGCSE Edexcel often asks you to describe the steps of micropropagation or adult cell cloning. Always use scientific terms: explant, agar, sterilise, callus, enucleation, electric shock, surrogate mother. Include a clear sequence. For plant cloning, mention why aseptic technique is important.

IGCSE Edexcel

For evaluation questions, give balanced arguments. Simply listing advantages will not earn full marks if the question asks you to ‘discuss’ or ‘evaluate’. Include a conclusion that weighs the benefits against the ethical or practical drawbacks. Suitable connectives: however, on the other hand, in contrast, therefore.

Remember that cloning does not increase variation; it reduces it. Cloning is often linked to selective breeding and genetic engineering topics, so be prepared to integrate knowledge across the specification.
