1. Protein Monomers: Amino Acids |

Proteins are polymers made up of monomers called amino acids. There are about 20 different amino acids commonly found in living organisms. Each amino acid has the same basic structure: a central carbon atom bonded to an amino group (–NH₂), a carboxyl group (–COOH), a hydrogen atom, and a variable ‘R’ group. The R group is what makes each amino acid unique.

2. Peptide Bonds and Polypeptides |

Amino acids are linked together by condensation reactions, which remove a water molecule. The bond formed between the carboxyl group of one amino acid and the amino group of another is called a peptide bond. A chain of amino acids is called a polypeptide. A protein consists of one or more polypeptide chains folded into a specific shape.

3. Protein Structure: Four Levels |

Proteins have a hierarchy of structure. The primary structure is the sequence of amino acids in the polypeptide chain. The secondary structure refers to local folding patterns like alpha helices and beta pleated sheets, held by hydrogen bonds. The tertiary structure is the overall 3D shape of a single polypeptide, stabilised by interactions between R groups — including hydrogen bonds, ionic bonds, and disulfide bridges. Quaternary structure exists when a protein has more than one polypeptide subunit, e.g., haemoglobin has four.

4. Importance of Shape for Function |

A protein’s function depends critically on its specific three-dimensional shape. This shape allows it to recognise and bind to other molecules (ligands). If the shape is altered — for example, through high temperature or extreme pH — the protein may no longer function. This is because the bonds holding the tertiary structure break, causing the protein to unfold. This process is called denaturation.

Denaturation is often irreversible. High temperatures break hydrogen bonds and ionic bonds in the protein, causing it to lose shape. Extreme pH disrupts ionic and hydrogen bonds by altering the charges on the R groups. Denatured enzymes lose their active site shape, so the substrate can no longer bind, stopping the reaction. In the exam, you may be asked to explain why an enzyme rate drops sharply above its optimum temperature.

6. Enzymes: Biological Catalysts |

Enzymes are globular proteins that speed up metabolic reactions by lowering the activation energy. They have a specific region called the active site, which is complementary in shape to the substrate. Enzyme action is explained by the lock and key model — the substrate fits exactly into the active site, like a key in a lock. An alternative model called the induced fit model is mentioned in some contexts, but the lock and key model is standard for Edexcel GCSE.

7. Factors Affecting Enzyme Activity |

Temperature and pH both affect enzyme activity. As temperature increases, kinetic energy of molecules rises, so collisions are more frequent, increasing the rate of reaction up to an optimum. Beyond this optimum, the enzyme denatures and the rate falls sharply. pH affects the charges on the enzyme’s active site; each enzyme has an optimum pH. Substrate concentration also affects rate: as concentration increases, rate increases until all active sites are occupied (saturation point).

8. Word and Chemical Equations for Digestion |

Many specification points require you to recall word equations for enzyme reactions, like starch → maltose (amylase), proteins → amino acids (protease), lipids → glycerol + fatty acids (lipase). For example: Starch + Water → (amylase) → Maltose. You may also be asked to write the balanced hydrolysis of a dipeptide, e.g., Dipeptide + H₂O → Amino acid 1 + Amino acid 2.

9. Core Practical: Effect of pH on Enzyme Activity | 核心实验:pH 对酶活性的影响

A common Edexcel core practical investigates how pH affects the rate of an enzyme-controlled reaction, often using amylase and starch. The method typically involves placing amylase solution and starch solution in separate test tubes at a set temperature, mixing them, and then taking timed samples to test with iodine solution. The time taken for the iodine to stop turning blue-black (indicating all starch has been broken down) is recorded. Rate = 1 / time taken. A graph of rate against pH shows a bell-shaped curve with an optimum.

10. Other Experiment Considerations |

Key variables to control: temperature (use a water bath), enzyme concentration, substrate concentration, and volume of solutions. The independent variable is pH (using buffer solutions). Safety: wear goggles; iodine is an irritant. Always measure the time until the iodine no longer turns blue-black. If the reaction happens too quickly, dilute the enzyme or reduce temperature. Repeat for reliability and calculate a mean rate.

自变量是 pH(使用缓冲液)。

11. Food Tests for Protein |

Edexcel GCSE requires the Biuret test for proteins. Add an equal volume of sodium hydroxide solution to the food sample, then add a few drops of dilute copper(II) sulfate solution. Mix gently. A purple or mauve colour indicates the presence of protein. A blue colour shows no protein. This is a qualitative test. You must know the reagents and the expected colour change.

12. Protein Functions and Examples |

Proteins have a huge range of functions. Structural proteins like collagen provide strength in skin and bones. Hormones such as insulin are proteins that regulate blood glucose. Haemoglobin is a transport protein carrying oxygen in red blood cells. Antibodies are Y-shaped proteins that defend against pathogens. Enzymes like catalase break down hydrogen peroxide. In the exam, you may be asked to link a protein’s shape to its job.
