Cellular Control: Gene Regulation and Expression |

📚 Cellular Control: Gene Regulation and Expression |

All living organisms need to control when and where their genes are expressed. This regulation underpins development, differentiation, and the ability to respond to environmental signals. For A‑level Biology, the key areas include prokaryotic gene control (the lac operon), eukaryotic transcription factors, epigenetic changes, RNA interference, and programmed cell death. Mastery of these topics is essential for tackling exam questions on genetic control and biotechnology.

1. Principles of Cellular Control |

Gene expression is regulated at multiple levels: transcriptional, post‑transcriptional, translational, and post‑translational. Transcriptional control is the most common and energy‑efficient point of regulation. Proteins such as transcription factors bind to specific DNA sequences to either promote or block the recruitment of RNA polymerase.

In prokaryotes, genes with related functions are often organised into operons and transcribed together as a single mRNA molecule. In eukaryotes, each gene usually has its own promoter and is regulated by a complex interplay of activator and repressor proteins, often acting over long distances.

2. Gene Mutations and Their Impact on Control |

Mutations in coding or regulatory sequences can severely disrupt cellular control. A single base substitution in a promoter may reduce the binding affinity of a transcription factor, while a frameshift mutation in a tumour suppressor gene can lead to unregulated cell division.

Gain‑of‑function mutations in proto‑oncogenes convert them into oncogenes, producing hyperactive proteins that constantly signal the cell to divide. Loss‑of‑function mutations in genes encoding DNA repair enzymes increase the mutation rate, further destabilising the genome.

Point mutations can create premature stop codons (nonsense) or alter active sites (missense), while insertions or deletions in control elements can remove entire transcription factor binding sites. Such changes are the molecular basis of many cancers.
