Introduction: Why Cellular Respiration Matters
Cellular respiration is one of the most fundamental biochemical processes in living organisms. It is the mechanism by which cells convert the chemical energy stored in organic molecules—primarily glucose—into ATP (adenosine triphosphate), the universal energy currency of the cell. For A-Level Biology students, understanding cellular respiration is not just about memorising a sequence of reactions; it is about appreciating how structure and function are intimately linked at the molecular level, from the compartmentalisation of the mitochondrion to the precise orientation of protein complexes in the inner membrane.
In this article, we will walk through the four major stages of aerobic respiration—glycolysis, the link reaction, the Krebs cycle, and oxidative phosphorylation—before examining anaerobic respiration and the overall ATP yield. Every step will be explained with clarity, linking biochemical detail to the broader physiological context that exam boards expect you to master.
1. Overview of Aerobic Respiration
The overall equation for aerobic respiration is deceptively simple:
C₆H₁₂O₆ + 6O₂ → 6CO₂ + 6H₂O + Energy (ATP)
However, this single equation conceals a sequence of over 20 enzyme-catalysed reactions distributed across the cytoplasm and the mitochondrion. The process can be divided into four main stages:
- Glycolysis – occurs in the cytoplasm; splits glucose into pyruvate and produces a small net gain of ATP and NADH.
- Link Reaction – occurs in the mitochondrial matrix; converts pyruvate into acetyl-CoA, releasing CO₂ and generating NADH.
- Krebs Cycle (Citric Acid Cycle) – occurs in the mitochondrial matrix; completes the oxidation of acetyl-CoA, producing ATP, NADH, FADH₂, and CO₂.
- Oxidative Phosphorylation – occurs across the inner mitochondrial membrane; uses the electron transport chain and chemiosmosis to generate the vast majority of ATP.
A key concept to grasp early is the role of electron carriers. NAD⁺ and FAD are coenzymes that accept electrons and protons during oxidation reactions, becoming reduced to NADH and FADH₂. These reduced coenzymes then shuttle their high-energy electrons to the electron transport chain, where the energy is finally harnessed to drive ATP synthesis. Think of NADH and FADH₂ as “charged batteries” carrying energy from the early stages to the final, most productive stage.
2. Glycolysis: The Universal First Step
Glycolysis (from the Greek glykys, “sweet”, and lysis, “splitting”) takes place in the cytoplasm and does not require oxygen—it occurs in both aerobic and anaerobic organisms. It is arguably the most ancient metabolic pathway, conserved across all domains of life.
Location: Cytoplasm
Input: 1 Glucose (6C)
Output: 2 Pyruvate (3C), net 2 ATP, 2 NADH
The process occurs in two phases: energy investment and energy payoff.
Energy Investment Phase (Phosphorylation of Glucose)
Glucose is phosphorylated twice using 2 ATP molecules. This serves two purposes: it traps glucose inside the cell (phosphorylated glucose cannot cross the plasma membrane via GLUT transporters), and it destabilises the molecule, making it more reactive for subsequent cleavage. The key enzyme here is phosphofructokinase (PFK), which catalyses the third step—the phosphorylation of fructose-6-phosphate to fructose-1,6-bisphosphate. PFK is a crucial regulatory enzyme; it is inhibited by high levels of ATP and citrate (feedback inhibition), and activated by AMP, ensuring glycolysis proceeds only when the cell needs energy.
The phosphorylated 6-carbon sugar is then split into two 3-carbon molecules: glyceraldehyde-3-phosphate (GALP) and dihydroxyacetone phosphate (DHAP), which is rapidly isomerised to GALP.
Energy Payoff Phase (Oxidation and ATP Formation)
Each GALP molecule undergoes oxidation, during which NAD⁺ is reduced to NADH. Subsequent reactions involve substrate-level phosphorylation: the direct transfer of a phosphate group from a phosphorylated intermediate to ADP, forming ATP. This is distinct from oxidative phosphorylation, which uses the proton gradient. For each glucose molecule entering glycolysis, 4 ATP are produced in this phase. Subtracting the 2 ATP invested earlier yields a net gain of 2 ATP per glucose.
The end product is pyruvate. In the presence of oxygen, pyruvate enters the mitochondrion for the link reaction. In the absence of oxygen, it is converted to lactate (in animals) or ethanol (in yeast and plants) via anaerobic respiration.
3. The Link Reaction: Bridging Glycolysis and the Krebs Cycle
Pyruvate cannot enter the Krebs cycle directly. It must first be converted to acetyl-CoA in a process called the link reaction (also known as pyruvate oxidation or the pyruvate dehydrogenase complex reaction).
Location: Mitochondrial Matrix
Input per pyruvate: 1 Pyruvate (3C) + CoA + NAD⁺
Output per pyruvate: 1 Acetyl-CoA (2C) + 1 CO₂ + 1 NADH
The reaction is catalysed by the pyruvate dehydrogenase complex, a massive multi-enzyme assembly. Pyruvate undergoes decarboxylation (loss of CO₂) and dehydrogenation (removal of hydrogen atoms, which are accepted by NAD⁺ to form NADH). The remaining 2-carbon acetyl group is then attached to coenzyme A (CoA), forming acetyl-CoA.
Since one glucose molecule yields two pyruvate molecules, the link reaction occurs twice per glucose, producing:
- 2 Acetyl-CoA (which enter the Krebs cycle)
- 2 CO₂ (released as waste)
- 2 NADH (proceed to oxidative phosphorylation)
Note that no ATP is produced directly in the link reaction—its value lies entirely in generating reduced NADH for the electron transport chain and in preparing the carbon skeleton for the Krebs cycle.
(3C)+ CoA + NAD⁺
- 2个乙酰辅酶A(进入克雷布斯循环)
- 2个NADH(进入氧化磷酸化)
4. The Krebs Cycle: The Metabolic Hub
Named after Sir Hans Krebs, who elucidated the cycle in 1937, the Krebs cycle (also called the citric acid cycle or tricarboxylic acid (TCA) cycle) is a closed loop of eight enzyme-catalysed reactions that completes the oxidation of acetyl-CoA to CO₂.
Location: Mitochondrial Matrix
Input per turn: 1 Acetyl-CoA (2C)
Output per turn: 3 NADH + 1 FADH₂ + 1 ATP (via GTP) + 2 CO₂
Per glucose (2 turns): 6 NADH + 2 FADH₂ + 2 ATP + 4 CO₂
The cycle can be summarised in four key transformations:
- Entry of acetyl-CoA: Acetyl-CoA (2C) combines with oxaloacetate (4C) to form citrate (6C), catalysed by citrate synthase. CoA is released and recycled.
- Decarboxylation steps: Citrate is isomerised to isocitrate, then undergoes two successive decarboxylation reactions—first to α-ketoglutarate (5C), then to succinyl-CoA (4C). Each decarboxylation releases one CO₂ and reduces one NAD⁺ to NADH.
- Substrate-level phosphorylation: Succinyl-CoA is converted to succinate, and the energy released drives the phosphorylation of GDP to GTP, which is then used to make ATP. This is the only ATP produced directly in the Krebs cycle.
- Regeneration of oxaloacetate: Succinate is oxidised to fumarate (producing FADH₂), then to malate, and finally to oxaloacetate (producing NADH). The regenerated oxaloacetate is ready to accept another acetyl-CoA.
The Krebs cycle is not just a catabolic pathway—it is amphibolic, meaning intermediates can be drawn off for biosynthesis (e.g., amino acids, fatty acids, haem groups) and replenished through anaplerotic reactions. This dual role makes it the central hub of cellular metabolism.
6 NADH + 2 FADH₂ + 2 ATP + 4 CO₂
5. Oxidative Phosphorylation: The Main Event
Oxidative phosphorylation is where the vast majority of ATP is produced—approximately 28 out of the 32 ATP molecules per glucose. It consists of two tightly coupled processes: the electron transport chain (ETC) and chemiosmosis.
Location: Inner Mitochondrial Membrane
The inner mitochondrial membrane is highly folded into cristae, dramatically increasing its surface area. Embedded within this membrane are four protein complexes (I–IV) and two mobile carriers (ubiquinone and cytochrome c) that constitute the electron transport chain, along with ATP synthase (sometimes called Complex V).
The Electron Transport Chain (ETC)
NADH and FADH₂, produced in earlier stages, donate their high-energy electrons to the ETC:
- Complex I (NADH dehydrogenase): NADH donates electrons, which are passed to ubiquinone (Q). Protons (H⁺) are pumped from the matrix into the intermembrane space.
- Complex II (Succinate dehydrogenase): FADH₂ donates electrons directly to ubiquinone. No protons are pumped here—this is why FADH₂ yields fewer ATP than NADH.
- Complex III (Cytochrome bc1 complex): Electrons from ubiquinol are transferred to cytochrome c. More protons are pumped across the membrane.
- Complex IV (Cytochrome c oxidase): Electrons are transferred from cytochrome c to the final electron acceptor—molecular oxygen (O₂)—which is reduced to water: ½O₂ + 2e⁻ + 2H⁺ → H₂O.
The sequential transfer of electrons through complexes of increasing electronegativity releases energy at each step. This energy is used to pump protons from the matrix into the intermembrane space, creating an electrochemical gradient (proton-motive force) across the inner membrane.
Chemiosmosis
The proton gradient established by the ETC represents stored potential energy. Protons can only flow back into the matrix through a specific channel: ATP synthase. As protons flow down their electrochemical gradient through ATP synthase (a process called chemiosmosis), the enzyme rotates, catalysing the phosphorylation of ADP to ATP:
ADP + Pi → ATP
This mechanism, proposed by Peter Mitchell in 1961 (for which he won the Nobel Prize in 1978), elegantly couples electron transport to ATP synthesis via a proton gradient—a principle now recognised as fundamental across all energy-transducing membranes.
ATP Yield per Reduced Coenzyme
The number of protons required to synthesise one ATP varies slightly, but the widely accepted estimates are:
- Each NADH → approximately 2.5 ATP
- Each FADH₂ → approximately 1.5 ATP
The difference arises because electrons from FADH₂ enter the ETC at Complex II, bypassing Complex I and thus pumping fewer protons.
ADP + Pi → ATP
6. Anaerobic Respiration: Life Without Oxygen
When oxygen is unavailable, the electron transport chain cannot function—there is no final electron acceptor to keep the chain flowing. Without a way to re-oxidise NADH back to NAD⁺, glycolysis would grind to a halt (it requires NAD⁺ for the oxidation of GALP). Anaerobic respiration solves this problem by regenerating NAD⁺ through alternative pathways.
Lactate Fermentation (Animals)
In animal cells (e.g., during vigorous exercise when oxygen delivery to muscles is insufficient), pyruvate is reduced to lactate by the enzyme lactate dehydrogenase. This reaction oxidises NADH back to NAD⁺, allowing glycolysis to continue producing 2 ATP per glucose.
Pyruvate + NADH → Lactate + NAD⁺
Lactate can be transported via the bloodstream to the liver, where it is converted back to glucose through the Cori cycle (gluconeogenesis). This is an energy-costly process, but it buys time for tissues when oxygen is limiting.
Ethanol Fermentation (Yeast and Plants)
In yeast and some plant tissues, pyruvate is first decarboxylated to ethanal (acetaldehyde), releasing CO₂, and then reduced to ethanol by alcohol dehydrogenase, regenerating NAD⁺.
Pyruvate → Ethanal + CO₂
Ethanal + NADH → Ethanol + NAD⁺
This process is commercially exploited in brewing and baking—the CO₂ released causes dough to rise, and the ethanol contributes to the flavour and preservation of alcoholic beverages.
The Efficiency Gap
Aerobic respiration yields approximately 32 ATP per glucose, while anaerobic respiration yields only 2 ATP. The ~16-fold difference underscores why oxygen is so critical for complex, energy-demanding organisms. However, anaerobic respiration is fast—it can produce ATP rapidly when demand outstrips oxygen supply, albeit with the accumulation of lactate as a metabolic cost.
7. ATP Yield Summary: Putting It All Together
A common exam question asks students to calculate the total ATP yield from the complete oxidation of one glucose molecule. The table below provides a clear breakdown:
| Stage | ATP (direct) | NADH | FADH₂ | ATP equiv. from NADH/FADH₂ |
|---|---|---|---|---|
| Glycolysis | 2 | 2 | 0 | 5 |
| Link Reaction (×2) | 0 | 2 | 0 | 5 |
| Krebs Cycle (×2) | 2 | 6 | 2 | 18 |
| TOTAL | 4 | 10 | 2 | ~32 ATP |
Notes: ATP from NADH ≈ 2.5; ATP from FADH₂ ≈ 1.5. Glycolysis occurs in the cytoplasm; its NADH may yield slightly less (≈1.5–2.5) depending on the shuttle used (malate-aspartate vs. glycerol-3-phosphate). The ~32 ATP total is a widely cited estimate; some textbooks give 30–38 depending on assumptions.
8. Key Exam Tips and Common Pitfalls
1. Know your locations. Exam boards love asking where each stage occurs. Glycolysis = cytoplasm. Link reaction, Krebs cycle = mitochondrial matrix. ETC and chemiosmosis = inner mitochondrial membrane (cristae).
2. Distinguish phosphorylation types. Substrate-level phosphorylation (direct transfer of phosphate to ADP) occurs in glycolysis and the Krebs cycle. Oxidative phosphorylation (proton gradient-driven) occurs at the inner membrane. Do not confuse the two.
3. Oxygen’s role. Oxygen is the final electron acceptor in the ETC—not a direct reactant in glycolysis or the Krebs cycle. Without O₂, the ETC backs up, NADH cannot be re-oxidised, and the entire aerobic pathway stalls.
4. Coenzyme counts per glucose. Remember to double everything after glycolysis: 2 pyruvate → 2 link reactions → 2 turns of the Krebs cycle. A common mistake is forgetting to multiply by 2.
5. Chemiosmosis requires an intact membrane. If the inner mitochondrial membrane is damaged, the proton gradient cannot be maintained, and oxidative phosphorylation fails—even if all enzymes are present.
6. Structure–function relationships. Be prepared to explain how mitochondrial structure supports its function: large surface area of cristae for ETC complexes and ATP synthase, intermembrane space for proton accumulation, matrix contains enzymes for link reaction and Krebs cycle.
Understanding cellular respiration at the depth required for A-Level is challenging but deeply rewarding. It connects disparate topics—enzyme action, membrane structure, transport mechanisms, and metabolic regulation—into a coherent whole. Master this topic, and you will have a template for understanding how cells harness energy, a skill that extends far beyond the exam hall.
