📚 AS Cambridge Biology: Core Knowledge Review | AS剑桥生物:核心知识点梳理

The AS Level Cambridge Biology syllabus (9700) lays the foundation for understanding life at the molecular, cellular, and systems level. This review covers the 11 core topics that every AS candidate must master, from the intricate structure of cells to the coordinated defence mechanisms of the immune system. Whether you are preparing for Paper 1 (multiple choice), Paper 2 (structured questions), or Paper 3 (practical skills), a solid grasp of these fundamentals is essential for success.

1. Cell Structure |

All living organisms are composed of cells, the basic structural and functional units of life. Cambridge AS Biology requires students to compare prokaryotic and eukaryotic cells in detail. Eukaryotic cells, found in animals, plants, fungi, and protoctists, possess a true nucleus enclosed by a nuclear envelope, along with membrane-bound organelles such as mitochondria, the endoplasmic reticulum, the Golgi apparatus, and lysosomes. Prokaryotic cells, such as bacteria, lack a nucleus and membrane-bound organelles; their genetic material exists as a circular DNA molecule in the nucleoid region, and they possess 70S ribosomes rather than the 80S ribosomes found in eukaryotes.

A key skill is the ability to identify organelles from electron micrographs and to relate structure to function. For example, the extensive cristae of mitochondria increase the surface area for oxidative phosphorylation, while the stacked thylakoids of chloroplasts maximise light absorption for photosynthesis. The relative sizes of organelles and cells should also be understood — students must be comfortable converting between millimetres, micrometres, and nanometres using the eyepiece graticule and stage micrometer calibration method.

Viruses occupy a unique position — they are non-cellular infectious particles consisting of nucleic acid (DNA or RNA) surrounded by a protein coat called a capsid. Since they lack cellular structure and cannot carry out metabolic processes independently, they are not classified as living organisms. Understanding viral structure is important for topics in infectious disease and immunity.

2. Biological Molecules |

Life is built from a relatively small number of chemical elements — primarily carbon, hydrogen, oxygen, nitrogen, phosphorus, and sulfur. These combine to form the four major groups of biological macromolecules: carbohydrates, lipids, proteins, and nucleic acids. Each group is composed of smaller monomeric units: monosaccharides for carbohydrates, fatty acids and glycerol for lipids, amino acids for proteins, and nucleotides for nucleic acids.

Carbohydrates serve as both energy sources and structural components. Glucose, a hexose monosaccharide with the formula C6H12O6, exists in two isomeric forms: alpha-glucose and beta-glucose. The orientation of the hydroxyl group on carbon-1 determines whether the polymer formed will be starch (from alpha-glucose), which is compact and ideal for energy storage in plants, or cellulose (from beta-glucose), which forms strong, straight chains suitable for cell wall construction. The glycosidic bonds linking monosaccharides — formed through condensation reactions and broken by hydrolysis — are a recurring theme throughout biochemistry.

Lipids are diverse hydrophobic molecules including triglycerides (fats and oils), phospholipids, and steroids. Triglycerides are formed by ester bonds between glycerol and three fatty acid chains. Their high ratio of carbon-hydrogen bonds to carbon-oxygen bonds makes them excellent long-term energy stores — gram for gram, lipids release approximately twice the energy of carbohydrates. The distinction between saturated and unsaturated fatty acids (presence or absence of carbon-carbon double bonds) determines the physical properties of fats and their health implications.

Proteins are the most structurally and functionally diverse macromolecules. They are polymers of amino acids joined by peptide bonds. AS students must know the general structure of an amino acid — an amino group (NH2), a carboxyl group (COOH), and a variable R group all attached to a central alpha carbon. The four levels of protein structure — primary (amino acid sequence), secondary (alpha-helices and beta-pleated sheets stabilised by hydrogen bonds), tertiary (3D folding maintained by hydrogen bonds, ionic bonds, hydrophobic interactions, and disulfide bridges), and quaternary (multiple polypeptide subunits) — determine a protein’s specific function. Haemoglobin and collagen are classic AS examples of quaternary structure.

3. Enzymes | 酶

Enzymes are biological catalysts — globular proteins that dramatically accelerate the rate of metabolic reactions without being consumed in the process. AS candidates must thoroughly understand the induced-fit model, where the binding of a substrate induces a conformational change in the enzyme’s active site, moulding it into a precise complementary shape. This contrasts with the older lock-and-key model, which assumes a rigid, pre-formed active site.

The factors affecting enzyme activity — temperature, pH, enzyme concentration, and substrate concentration — are central to the AS syllabus. Students must be able to interpret the characteristic bell-shaped curves of temperature and pH against reaction rate: low temperatures reduce kinetic energy and collision frequency; temperatures above the optimum cause denaturation as hydrogen bonds in the tertiary structure break; extreme pH values alter the ionisation of R groups in the active site, disrupting substrate binding. The concept of Vmax (maximum rate when all active sites are occupied) and the Michaelis-Menten constant (Km) is introduced at AS level as a way to quantify enzyme affinity for its substrate.

Enzyme inhibition is classified as competitive or non-competitive. Competitive inhibitors resemble the substrate and bind reversibly to the active site, an effect that can be overcome by increasing substrate concentration. Non-competitive inhibitors bind to an allosteric site, altering the shape of the active site without directly blocking it; increasing substrate concentration cannot reverse this inhibition. This distinction is frequently tested in AS exam questions, often with graphs showing Vmax and Km changes.

4. Cell Membranes and Transport |

The fluid mosaic model describes the structure of cell membranes: a phospholipid bilayer with hydrophobic fatty acid tails facing inward and hydrophilic phosphate heads facing outward. Embedded within this bilayer are proteins — integral proteins spanning the membrane and peripheral proteins attached to the surface — along with cholesterol molecules (in animal cells) that regulate membrane fluidity, and glycoproteins and glycolipids involved in cell recognition and signalling.

Transport across membranes occurs through several mechanisms. Simple diffusion is the passive net movement of small, non-polar molecules (such as oxygen and carbon dioxide) down their concentration gradient. Facilitated diffusion uses channel proteins (for ions and water) or carrier proteins (for larger polar molecules like glucose) — still passive but protein-mediated. Osmosis is the specific case of water diffusing through a partially permeable membrane from a region of higher water potential (less negative) to a region of lower water potential (more negative). Active transport requires metabolic energy (ATP) to move substances against their concentration gradient via specific carrier proteins, such as the sodium-potassium pump. Endocytosis and exocytosis are bulk transport mechanisms involving vesicle formation and fusion.

5. The Mitotic Cell Cycle |

The cell cycle is a precisely regulated sequence of events by which a cell duplicates its contents and divides into two daughter cells. It consists of interphase (G1, S, and G2 phases) followed by mitosis and cytokinesis. During interphase, the cell grows, carries out normal metabolic activities, and replicates its DNA. The S (synthesis) phase is critical — each chromosome is duplicated to form two identical sister chromatids held together at the centromere.

Mitosis is divided into four stages for examination purposes: prophase (chromosomes condense and become visible, nuclear envelope breaks down, spindle fibres form), metaphase (chromosomes align at the metaphase plate equator, spindle fibres attach to centromeres), anaphase (centromeres split, sister chromatids are pulled to opposite poles by shortening spindle fibres), and telophase (chromosomes decondense, nuclear envelopes reform around each set). Cytokinesis follows, dividing the cytoplasm; in animal cells this occurs through a cleavage furrow, while in plant cells a cell plate forms.

Understanding the role of mitosis in growth, repair of tissues, and asexual reproduction is essential. AS students should also be introduced to the concept that cancer results from uncontrolled cell division due to mutations in genes that regulate the cell cycle, such as proto-oncogenes and tumour suppressor genes.

6. Nucleic Acids and Protein Synthesis |

Nucleic acids — DNA and RNA — are polymers of nucleotides, each consisting of a pentose sugar, a phosphate group, and a nitrogenous base. DNA is a double helix of two antiparallel polynucleotide strands held together by hydrogen bonds between complementary base pairs: adenine (A) pairs with thymine (T) via two hydrogen bonds, and cytosine (C) pairs with guanine (G) via three hydrogen bonds. RNA is typically single-stranded and contains ribose instead of deoxyribose, with uracil (U) replacing thymine.

DNA replication is semi-conservative — each new DNA molecule consists of one original (parental) strand and one newly synthesised strand. The Meselson and Stahl experiment, which used heavy nitrogen (15N) isotopes and density-gradient centrifugation, provided elegant experimental evidence for this model. The enzyme DNA helicase unwinds the double helix, while DNA polymerase adds complementary nucleotides to the template strand in the 5′ to 3′ direction, forming a continuous leading strand and a discontinuous lagging strand composed of Okazaki fragments joined by DNA ligase.

Protein synthesis involves two major stages: transcription and translation. During transcription, the enzyme RNA polymerase binds to a promoter region, unwinds a section of DNA, and synthesises a complementary messenger RNA (mRNA) strand. In eukaryotes, the pre-mRNA undergoes splicing — introns are removed and exons are joined — before the mature mRNA exits the nucleus. Translation occurs on ribosomes in the cytoplasm: the mRNA sequence is read in triplet codons, each specifying a particular amino acid. Transfer RNA (tRNA) molecules with complementary anticodons deliver the appropriate amino acids, and peptide bonds form between adjacent amino acids to build the polypeptide chain. The genetic code is universal, degenerate, and non-overlapping — key features that AS students must be prepared to explain.

7. Transport in Plants |

Plants require efficient transport systems to move water, minerals, and the products of photosynthesis throughout the organism. The xylem and phloem are the two vascular tissues that form the plant transport system. Xylem vessels transport water and dissolved mineral ions from the roots to the leaves. Xylem vessel elements are dead at maturity, with their end walls broken down to form continuous hollow tubes. Their walls are strengthened with lignin, which provides structural support and prevents collapse under the tension created by transpiration.

The cohesion-tension theory explains water movement in the xylem. Transpiration from leaf mesophyll cells lowers water potential, creating a tension that pulls water up the xylem. Water molecules cohere to each other through hydrogen bonding, maintaining an unbroken column of water from roots to leaves. The rate of transpiration is affected by temperature, humidity, wind speed, and light intensity — students should be able to design and interpret experiments using a potometer to measure transpiration rates.

Phloem tissue transports assimilates, principally sucrose and amino acids, from sources (where they are produced or stored, such as mature leaves) to sinks (where they are used or stored, such as roots, developing fruits, and meristems). The mass flow hypothesis, also called the pressure-flow model, proposes that active loading of sucrose into the sieve tube elements at the source lowers water potential, causing water to enter by osmosis and creating high hydrostatic pressure. At the sink, sucrose is unloaded, water leaves, and the pressure drops. The resulting pressure gradient drives the bulk flow of phloem sap from source to sink. Companion cells provide metabolic support for the enucleate sieve tube elements, including the ATP required for active loading.

8. Transport in Mammals: The Circulatory System |

The mammalian circulatory system is a closed, double circulatory system. The heart is a muscular organ with four chambers: two thin-walled atria that receive blood and two thick-walled ventricles that pump blood out. The right side of the heart pumps deoxygenated blood to the lungs via the pulmonary artery (pulmonary circulation), while the left side pumps oxygenated blood to the rest of the body via the aorta (systemic circulation). The left ventricle has a thicker muscular wall than the right because it must generate sufficient pressure to propel blood throughout the entire body.

The cardiac cycle consists of systole (contraction) and diastole (relaxation). The sinoatrial node (SAN) in the right atrium acts as the heart’s natural pacemaker, generating electrical impulses that spread across the atria, causing atrial systole. The impulse is then delayed at the atrioventricular node (AVN) before travelling down the bundle of His and Purkinje fibres to trigger ventricular systole from the apex upwards. Understanding this sequence and the associated pressure changes in the atria, ventricles, and major arteries is essential for interpreting electrocardiogram (ECG) traces.

Blood vessels are specialised for their roles in circulation. Arteries carry blood away from the heart under high pressure; their walls contain thick layers of smooth muscle and elastic tissue that allow them to stretch and recoil, maintaining pressure during diastole. Veins return blood to the heart under low pressure; they have thinner walls, wider lumens, and valves to prevent backflow. Capillaries are the site of material exchange between blood and tissues; their walls consist of a single layer of endothelial cells, providing a short diffusion distance. AS students should relate the structure of each vessel type to its function and be able to recognise them from photomicrographs.

9. Gas Exchange and Smoking |

The human gas exchange system is adapted for the efficient uptake of oxygen and removal of carbon dioxide. Air enters through the nasal passages and trachea, passes through the bronchi and bronchioles, and reaches the alveoli — millions of tiny air sacs where gas exchange occurs. The alveoli are ideally suited for their function: they provide a large total surface area (approximately 70 m2 in an adult), a thin diffusion barrier (the alveolar and capillary walls are each just one cell thick), a steep concentration gradient maintained by continuous blood flow and ventilation, and a moist surface for the dissolution of gases.

AS students should understand the mechanism of ventilation. During inspiration, the external intercostal muscles contract (raising the rib cage) and the diaphragm contracts (flattening), increasing the volume of the thoracic cavity and decreasing its pressure below atmospheric pressure, so air flows in. During expiration at rest, these muscles relax passively; the elastic recoil of the lungs and chest wall reduces thoracic volume, increasing pressure and forcing air out. Forced expiration involves contraction of the internal intercostal muscles and abdominal muscles.

The link between smoking and disease is a significant component of the syllabus. Tar, a component of cigarette smoke, paralyses and destroys cilia in the airways, impairing the mucociliary escalator and leading to chronic bronchitis. It is also the primary carcinogen responsible for lung cancer. Carbon monoxide binds irreversibly to haemoglobin with an affinity approximately 250 times greater than oxygen, reducing the oxygen-carrying capacity of blood. Nicotine is a vasoconstrictor that increases heart rate and blood pressure, contributing to cardiovascular disease and increasing the risk of thrombosis.

10. Infectious Diseases |

Infectious diseases are caused by pathogens — organisms that live in or on a host organism, causing harm. The four main types of pathogens covered at AS level are bacteria, viruses, fungi, and protoctists (protists). AS students are expected to study specific examples: cholera (caused by the bacterium Vibrio cholerae), tuberculosis (caused by Mycobacterium tuberculosis), malaria (caused by protoctists of the genus Plasmodium), and HIV/AIDS (caused by the human immunodeficiency virus).

For each disease, students must know the causative organism, its mode of transmission, the symptoms and effects on the body, and methods of prevention and control. Cholera is transmitted via faecally contaminated water and food; the cholera toxin causes chloride ion channels in intestinal epithelial cells to open, leading to massive water loss through osmosis and potentially fatal dehydration. Tuberculosis is an airborne disease spread by droplet infection. Malaria is vector-borne, transmitted by female Anopheles mosquitoes. HIV is transmitted through the exchange of bodily fluids and attacks helper T cells, progressively destroying the immune system.

Antibiotics are a major weapon against bacterial infections. They work by targeting features specific to prokaryotic cells — such as bacterial cell wall synthesis (penicillin), protein synthesis (tetracycline), or nucleic acid synthesis — without harming eukaryotic host cells. The emergence of antibiotic resistance, driven by natural selection when antibiotics are overused or misused, is a growing global health concern that AS students should be prepared to discuss, including strategies such as completing prescribed courses and reducing unnecessary prescriptions.

11. Immunity | 免疫

The immune system provides defence against pathogens through a coordinated network of cells, tissues, and molecules. AS students must distinguish between non-specific (innate) and specific (adaptive) defences. Innate defences are present from birth and respond rapidly: physical barriers such as skin and mucous membranes, phagocytosis by neutrophils and macrophages, inflammation, and the action of natural killer cells. These responses do not differentiate between different pathogens.

Specific immune responses involve lymphocytes (B cells and T cells) and are characterised by specificity and immunological memory. B lymphocytes produce antibodies — Y-shaped proteins (immunoglobulins) with variable regions that bind specifically to antigens on the pathogen surface. This is the humoral response. T lymphocytes are responsible for cell-mediated immunity: helper T cells release cytokines that activate B cells and cytotoxic T cells, while cytotoxic T cells directly destroy infected body cells displaying foreign antigens on their MHC class I molecules.

Vaccination exploits the principle of immunological memory. By introducing a harmless form of an antigen (attenuated pathogen, killed pathogen, or antigenic fragment), the primary immune response is triggered, generating memory B and T cells. Upon subsequent exposure to the actual pathogen, the secondary response is faster, stronger, and more prolonged, often preventing disease entirely. The concept of herd immunity — where a sufficiently high proportion of a population is immunised to protect vulnerable individuals — is a key application of this principle in public health policy.

12. Exam Tips for AS Cambridge Biology | AS剑桥生物考试技巧

Success in AS Cambridge Biology requires more than factual recall — it demands the ability to apply knowledge to unfamiliar contexts, analyse data, and communicate scientific ideas precisely. Here are some key strategies for each paper type.

Paper 1 (Multiple Choice): Work systematically through the 40 questions in 60 minutes. Read every option carefully — Cambridge examiners often include distractors that are partially correct but not the best answer. Pay special attention to negatively phrased questions (“Which of the following is NOT…”). If unsure, eliminate obviously wrong options first, then make an educated guess from the remainder. There is no penalty for incorrect answers.

Paper 2 (Structured Questions): Command words are critical. “Describe” requires stating what is observed without explanation; “Explain” demands reasons and mechanisms; “Suggest” invites you to apply knowledge to a new scenario. Note the mark allocation — a 3-mark question typically expects three distinct points. Use correct scientific terminology at all times; marks are regularly lost for vague or colloquial language. For graph questions, always read axis labels and units carefully before answering.

Describe(描述)”要求陈述所观察到的情况而不加解释;”Explain(解释)”要求给出原因和机制;”Suggest(建议)”邀请你将知识应用于新情境。

Paper 3 (Practical Skills): This paper tests experimental design, data handling, and evaluative skills. When asked to plan an investigation, clearly state the independent, dependent, and control variables. Describe how you would standardise control variables and ensure reliable results (e.g., replicates, large sample sizes). When evaluating data or experimental procedures, identify sources of error, suggest realistic improvements, and comment on the reliability and validity of conclusions. Practise drawing clear, well-labelled graphs with appropriate scales.

Mastering the AS Cambridge Biology syllabus is an ambitious undertaking, but a systematic approach to revision — actively testing yourself, drawing annotated diagrams, and practising past paper questions under timed conditions — will build both confidence and competence. Each of the eleven topics connects to the others in meaningful ways; recognising these interconnections is the hallmark of a high-achieving biology student.
